Out of curiosity I wanted to check the entry for "create manifest" in ASE "Commands" Reference manual. Normally caveats for using a command are explicitly mentioned in the "Command" manual.
"create manifest" is missing from "Commands" Reference manual versions 15.7 as well the latest 16.0.

From whatever I could read from mount, unmount and quiesce commands, the create manifest is trying to create and gather a full picture of a database for transport/load on a target.
For this to happen it may need exclusive locks on system tables related to a database namely sysdevices, sysusages, etc. So my hunch is you can create only one manifest for a database at a time. You should be able to check if another "create manifest" is in progress by searching MDA tables.
A similar locking on system table happens for example when you try alter database to expand and dump that database run at the same time.
